Description
Recent studies have found that genetic factors are the main reason for individual differences in warfarin dosage. The VKORC1 gene polymorphism related to the mechanism of action of warfarin and the CYP2C9 gene related to warfarin metabolism are closely related to its anticoagulant efficacy. Common variants that reduce the metabolic rate of warfarin are CYP2C9*2 and CYP2C9*3. In vivo and in vitro studies have shown that CYP2C9*2 and CYP2C9*3 impair the metabolism of S-warfarin by about 30-40% and about 80-90%, respectively.
The mutation of the gene site (-1639G>A) on the VKORC1 promoter is also closely related to the dose of warfarin.
