Description
The TWIST1 gene encodes a key helix-loop-helix (bHLH) transcription factor that plays a central role in embryonic development, particularly mesoderm formation and cell fate determination. However, its aberrant expression in adults is closely linked to tumorigenesis and progression. TWIST1 is a key driver of epithelial-mesenchymal transition (EMT), a process that enables epithelial cells to acquire mesenchymal characteristics, enhancing their ability to migrate, invade, and resist apoptosis, ultimately promoting cancer invasion, metastasis, and chemoresistance. Consequently, TWIST1 is frequently overexpressed in various malignancies (such as breast, gastric, and colorectal cancers) and is considered a key oncogene and a marker of poor prognosis.
The expression of the TWIST1 gene is tightly regulated by the DNA methylation status of its promoter region. In normal tissues, its promoter is typically hypermethylated, which represses gene transcription and silences its expression. However, in many cancers, this region undergoes demethylation (or hypomethylation), resulting in the release of transcriptional repression and aberrant activation and overexpression of the TWIST1 gene.
The TWIST1 methylation standard is a research tool designed to target this epigenetic alteration. It consists of a precisely quantified DNA fragment treated with bisulfite in vitro. The CpG sites in the TWIST1 gene promoter region are pre-set to a fully methylated state. It serves as an essential positive control and quantitative standard in assays such as MSP (methylation-specific PCR), BSP (bisulfite sequencing PCR), or pyrosequencing. Its primary value lies in: 1) verifying the validity of the experimental system, ensuring complete bisulfite conversion and specific PCR amplification; and 2) generating a standard curve to enable absolute quantification of TWIST1 methylation levels in clinical samples, thereby enabling precise and standardized early cancer diagnosis, prognostic assessment, and ongoing monitoring.
