Description
The protein encoded by the PTGER4 (prostaglandin E receptor 4, also known as EP4) gene is one of the receptors for prostaglandin E2 (PGE2). PGE2 is a lipid mediator found ubiquitously in the body and plays a key role in inflammation, immunity, and tumorigenesis.
Normal Function: Activation of the PTGER4 protein by PGE2 triggers a series of intracellular signaling pathways that regulate processes such as cell proliferation, apoptosis, angiogenesis, and immune responses.
Relationship with Cancer: PGE2 levels are abnormally elevated in various cancers. Sustained activation of the PTGER4 signaling pathway promotes tumor growth, invasion, and metastasis, while also helping tumor cells evade immune surveillance. Therefore, PTGER4 itself is often considered a driver of cancer risk.
However, aberrant gene methylation often leads to its silencing and inactivation. Hypermethylation of the PTGER4 gene promoter region has been most extensively studied in colorectal cancer (CRC).
As a biomarker: During the development and progression of colorectal cancer, the promoter region of the PTGER4 gene becomes hypermethylated, resulting in the inhibition of its transcription. This aberrant methylation occurs early in the progression of cancer and can be detected in patient stool or blood samples. Therefore, the methylation status of the PTGER4 gene (rather than the gene itself) has been developed as a highly promising biomarker for the noninvasive diagnosis and early screening of colorectal cancer.
Other cancers: In addition to colorectal cancer, aberrant methylation of the PTGER4 gene has also been found in malignancies such as gastric cancer and lung cancer. However, its primary and core application scenario is currently focused on the screening and diagnosis of colorectal cancer.
The PTGER4 methylation standard is a quality control tool used to monitor the quality of PTGER4 gene methylation testing. While the PTGER4 gene itself is a factor that promotes cancer development, hypermethylation of its promoter is a key marker for the development of cancers such as colorectal cancer. Measuring its methylation levels, particularly through fecal DNA testing, has become an important non-invasive early screening tool for colorectal cancer.
